It's really just up to the author. I have read novels where omegas still had heats while pregnant. So it just boils down to what the author decides

Because he took the drug, he was told his heat would be irregular. As for why it would happen while pregnant, it is up to the author but pregnancy is a hormonal surge; a majority of women are horny while pregnant with many factors as to why they don't act upon it. Probably what's meant to be outside of the drug screwing that up.
I guess I meant the way that heat/estrus is meant to attract a partner so that you can conceive, so the body doing it again while already pregnant just didn't make sense.
I completely forgot about the drug thing happening. If it's just being used as a 'horny must fuck' plot device then I guess the logistics don't really gotta makes sense.
How is he in heat if he's pregnant?? The biology is not biology-ing.
