fairly a new thing, the best example i can give is that it's like omegaverse and its whole purpose is like omegaverse - it lays down the groundwork to make the mc and mls have some sort of deeper connection and a reason to prove they're "made for each other" by "nature."
guideverse is a world where there are espers, who have different type of powers that we would typically see in normal shounen dungeon manhwas, and they fight monsters from those dungeons or something like that. when these espers use their powers too much, they have something like a breakdown, i don't remember the typical name they use in manhwas, but it makes them rogue, their sanity breaks, and they might/will kill others while they're in this rogue state, and eventually self combust/die.
this is where the guide's role comes in, they're not espers, they're their own seperate thing. their role is to guide these espers and calm them down in a typical bl fashion sense - by physical touch 99.9999% of the time. it's sometimes enough to hold hands, but ofc this is a bl and it leads to sex most of the time anyways.
some manhwas delve deeper into how the guide's powers work, like they send out some brainwaves that calm the espers down. others not so much. it's not completely the typical healing powers we know of, they're like their own thing.
a core thing in guideverse is that the espers and guides have a compatibility rate by percentage, the higher the percentage, the better compatibility and the better the guiding is. every single story of guideverse i've read, the mcs and mls have top compatibility rate and it's usually the core reason why they get involved with each other. omegaverse has that soulmate omega-alpha thing, guideverse has compatibility rate. most ukes are guides, most espers are semes in guideverse.
Guideverse is a Hunter/Dungeon universe where there are "awakened" people with special powers. They are ranked F-class to S-class (lowest to highest).
Espers are the awakened individuals who have attack related skills. But to using their skills corrupts their mind, and over time they can go beserk or on a rampage. They lose control of their powers and are a major threat of basically exploding and damaging whole buildings (even whole towns if they're higher classed.)
So like yin and yang, the presence of Espers brought along Guides. Guides are the answer to Espers' problem. They're "healers" able to purify the corruption plaguing Espers when they use a lot of power, stopping them from going beserk and exploding. The purification needs physical contact - the more contact, the more effective the guiding (so of course a lot of smut comics use this to their advantage like they do with omegaverse forced sexual encounters).
Espers and Guides have Class Ranks as well as Compatibility. So a mid ranked C-Class Guide may have high compatibility with a high ranked S-Class Esper (C-Class with 90% Compatibility is better than the guiding from an A-class with 50% Comp.) Bad compatibility feels nauseating for Espers, so there are stories of Espers refusing guiding until they find their perfect person.
Then, much like Omegaverse, there's imprinting. That seals the relationship between 1 Guide and 1 Esper - neither can guide or receive guiding from anyone else other than their paired partner, which in this universe, seals their fate. The Esper has to protect their guide with their life. If their imprinted guide dies, they'll eventually go beserk and die.
Then there is also "forced guiding" and "reverse guiding". Forced is pretty much an Esper assaulting an unwilling Guide. And Reverse is the backlash from an Esper rejecting guiding, and it ends up hurting the guide when when the power recoils.

I was looking on Reddit and saw a comment mention how Guideverse is only Guide MCs, and that is so true. Omegaverse has stories where the other genders can share the MC spot and it's not just the omega as the leading character. So are there any Guideverse with Esper MC, or shared lead status? I wanna see the other POVs.
