He was already a target since he got named the boss mate ( in the beginning chapters) and the brother himself is judging the boss for his title not who he is. I'm pretty sure the boss is trying to go straight in business.
I don't think he knows he's the head of the organization just that he's a gangster. That whole meet a greet at the house shows control not understanding. The fact he's more angry at the fact that he's a gangster and not that he's a guy says something. BTW what does the boss organization deal in? ( I don't remember). So he's judging the person for his title not who he is as a person.
