Try "Higouhou Junai" by Fuwa Shinri. The main/1st couple's story is like that. It's not like one of those typical yaoi where the seme immediately falls for the uke after finding about the uke's feelings. My heart wrenched for the uke several times in this manga. There's also a side story about the seme's assistant/secretary(a glasses blond guy. Sorry, can't remember his name exactly) which seems to fit your 2nd requirement.

Really heart wrenching BL about unrequited love? I'd prefer if the couple were more on the adultish side (so like college), but that isn't necessary.
Oh and can the love please be requited at the end?
